About Stuart Agar Gel
- Sodium glycerophosphate acts as a buffer to maintain the pH of the medium.
- Sodium thioglycollate provides a reduced environment favorable for the survival of anaerobes.
- Methylene blue serves as a redox indicator - the medium is initially colorless indicating a reduced state, and turns blue in the presence of oxygen.
- Agar provides a semi-solid gel matrix to prevent desiccation of the specimen during transport.
- Demineralized water is used as the solvent.
- Calcium chloride helps maintain the permeability of the cell membranes of bacteria.

【Expected Cultivation Experiment】
The bacterial strains used for quality control of Stuart Transport Medium include:
Streptococcus pyogenes (ATCC 19615):
- Aerobic gram-positive coccus
- Quality control requirement: Able to be isolated again after aerobic subculture for 24 hours
Bacteroides fragilis (ATCC 25285):
- Obligate anaerobe
- Quality control requirement: Able to be isolated again after aerobic subculture for 24 hours
Pseudomonas aeruginosa (ATCC 27853):
- Aerobic gram-negative bacillus with weak oxygen requirement
- Quality control requirement: Able to be isolated again after aerobic subculture for 24 hours
The rationale for choosing these 3 strains is to represent gram-positive cocci, gram-negative bacilli, and anaerobes with different oxygen requirements, covering the typical application range of the medium comprehensively as appropriate quality control strains.
